槐芩软膏介导 NF-kB 信号通路对小鼠痔疮模型肛缘水肿及 创面愈合的影响

Effect of Huaiqin Ointment on perianal edema and wound healing in mouse model with hemorrhoids via NF-kB signal pathway

摘要: 目的 探讨槐芩软膏对小鼠痔疮模型肛缘水肿及创面愈合的疗效及机制。 方法 将 30 只 ICR 小鼠通过肛周局部注射乙酸法建立痔术后创面损伤模型,然后将小鼠随机分成对照组和治疗组。 造模第 2 天对照组小鼠给予低浓度高锰酸钾溶液清洗,治疗组小鼠给予低浓度高锰酸钾清洗复合涂抹槐芩软膏。 治疗 7 d 后,对小鼠肛缘肿胀程度及愈合面积进行统计;采用 ELISA 和 qPCR 法检测小鼠血浆或水肿组织中超敏 C 反应蛋白( hs-CRP) 、白细胞介素 6( IL-6) 、肿瘤坏死因子 α( TNF-α) 、血红蛋白水平;采用 HE 染色和免疫荧光检测肛缘组织中病理变化及炎性细胞浸润情况;采用 Western blot 检测水肿组织中核因子 kB 抑制蛋白 α( IkBα) 磷酸化水平及核内核因子 kB( NFkB) p65 含量。 结果 与对照组小鼠相比,治疗组小鼠肛缘肿胀评分及创面面积显著改善,血浆或肛缘组织 hsCRP、IL-6、TNF-α、血红蛋白水平显著下降(P<0. 05) 。 HE 染色和免疫荧光显示治疗组小鼠肛缘组织中炎性细胞浸润相比于对照组显著减少(P<0. 05) 。 Western blot 显示治疗组组织 IkBα 磷酸化及细胞核内 NF-kB p65 水平相比对照组显著降低(P<0. 05) 。 结论 槐芩软膏治疗改善小鼠痔疮模型肛缘组织炎性反应及水肿,促进创面愈合,其分子机制可能与 NF-kB 信号通路有关。

Abstract: Objective To explore the effect and potential mechanism of Huaiqin ointment on perianal edema and wound healing in mouse model with hemorrhoids. Method 30 ICR mice were used to establish the mouse model with hemorrhoids via injecting acetic acid to perianal region, then mice were divided into control group and treatment group randomly. Mice in control group were treated with low concentration potassium permanganate cleaning, and mice in treatment were treated with low concentration potassium permanganate cleaning plus applying Huaiqin ointment. After 7 days treatment, the degree of perianal edema and wound healing in perianal region was analyzed. The hs-CRP, IL-6, TNF-α, hemoglobin levels in plasma or perianal tissues were measured by ELISA or qPCR. HE staining and immunofluorescence were used to detect the inflammatory cell infiltration in perianal tissues. Western blot was used to measure the phosphorylation of IkBα and intranuclear NF-kB p65 contents in perianal tissues. Result The degree of perianal edema, wound healing, the blood hs-CRP, the cytokines ( IL-6, TNF-α) in blood or perianal tissues, hemoglobin in perianal tissues of treatment group ameliorated significantly compared to control (P<0. 05) . HE staining and immunofluorescence showed that the inflammatory cell infiltration in perianal tissues in treatment group was significantly fewer than control group ( P<0. 05) . Western blot showed that the IkBα phosphorylation and content of p65 in nuclear of treatment group was also significantly lower than control ( P < 0. 05 ) . Conclusion Huaiqin Ointment treatment attenuates inflammation response and perianal edema, promotes wound healing, of which mechanism is involved in NF-kB signal pathway.
